NUCLEAR FACTOR KAPPA B (NF-κB) ACTIVITY ACCORDING TO GRADE OF KNEE OSTEOARTHRITIS PATIENTS
Abstarct: We aimed to determine in this study the role of the transcription factor NF-κB which were thought to be a therapeutic target in osteoarthritis (OA) development. In this study, blood samples were collected from 100 patients with a diagnosis of OA and 50 healthy subjects. In these samples peripheral mononuclear blood cells (PBMCs) were extracted by standard Ficoll density-gradient centrifugation. NF-κB activation was quantified by way of enzyme-linked immunosorbent assay (ELISA) using the NF-κB kit according to the manufacturer's instructions. When the NF-κB activity is considered between patients and the control group, there is an increase in patients compared to the control group (p=0.405). At the same time, it has been determined that increased by gender (p=0.260; p=0.610). But these increases were not statistically significant. When we classify OA patients according to Kellgren-Lawrence (K-L) grading system, we were not found statistically significant difference between NF-κB activity (p=0.229). But the value of NF-κB activity was significantly higher at early stage (grade 1) compared to control and other stages. We are based on, inflammatory processes may contribute to the pathogenesis of OA patients, which were classified according to grade, but were not found difference between NF-κB activities in the patients groups.
