Fusarik Asit ve Enniatin-A Mikotoksinlerinin İnsan Umblikal Ven Endotel Hücreleri (HUVEC) Üzerine Sitotoksik Etkilerinin Değerlendirilmesi

Öz


Fusarik asit (FA) ve Enniatin-A (EN-A) Fusarium spp. cinsi mikotoksinler olup gıdaları ve yemleri kontamine edebilirler. Tahıllarda mikotoksin varlığı önemli sağlık sorunlarına yol açmakla birlikte ekonomik kayıplara da sebep olmaktadır. Bu çalışmada FA ve EN-A’nın potansiyel sitotoksik etkileri insan umblikal ven endotel (HUVEC) hücre hattında 3-(4,5-dimethylthiazolyl-2)-2,5 diphenyltetrazolium bromid (MTT) testi ile incelenmiştir. Her iki mikotoksinin de farklı konsantrasyonları (FA için 0.78, 1.56, 3.125, 6.25, 12.5, 25, 50, 100, 150, 200, 400 µg/mL, EN-A için 0.048, 0.098, 0.195, 0.39, 0.78, 1.56, 3.125, 6.25 µg/mL) 24 ve 48 saat muamele sürelerinde uygulanmıştır. Sonuç olarak, FA ve EN-A’nın hücre canlılığını (%), her iki muamele süresinde de yüksek konsantrasyonlarda önemli düzeyde düşürdüğü belirlenmiştir. FA’nın 24 saatlik uygulamasında IC50 (hücrelerin %50’sini öldüren) değerinin 150 μg/mL olduğu, 48 saatlik uygulamasında ise 200 μg/mL olduğu tespit edilmiştir. EN-A’nın ise IC50 değerinin 24 saatlik uygulamada 3.125 μg/mL, 48 saatlik uygulamada ise 1.56 μg/mL olduğu saptanmıştır. Bu sonuçlara göre FA ve EN-A’nın yüksek konsantrasyonlarda HUVEC hücrelerinde sitotoksik etki gösterdikleri gözlenmiştir. Ayrıca, EN-A mikotoksininin FA’dan daha fazla toksik etkiye sahip olduğu söylenebilir. Bu sitotoksik etki, hücre döngüsünde ve hücre proliferasyonunda yavaşlamaya  dolayısıyla DNA replikasyonun bozulmasına ve hücrelerde apoptozise neden olabilir.
